SUMMARY:
By this point, this has kind of been superseded by Pack 7. But if it's on sale, or Party Pack 7 isn't for you, this is a good buy. Has a couple of the best Jackbox games, one that's kind of a dud IMO, and one that is good but can only be played IRL.
--------------------
TRIVIA MURDER PARTY
A standard trivia game with some minigames to break up the action.
Trivia games are not my thing, but the minigames are neat. A sequel was made for Party Pack 6, but the rest of the games on that pack aren't the greatest, so if you really like trivia games you might consider getting this pack instead.
---
QUIPLASH 2
You get a prompt. Another player gets the same prompt. Everyone else votes on which of you had the funnier answer.
Jackbox at its purest and simplest. Has a sequel in Party Pack 7, which has a much better final round. But there are some good prompts here that aren't in 3, so it's worth having both.
---
GUESSPIONAGE
You get a survey question, and you have to guess the percentage of people who answered a certain way to the question. Everyone else guesses if you're too high or too low.
Mildly interesting, but not much more. Remember the Everybody Votes Channel from the Wii? It's like that, but as a Jackbox game.
---
TEE K.O.
You draw a few things, you write a few phrases, and those drawings and phrases are sent off to everyone else. They match drawings with phrases to make a funny T-shirt.
Quite possibly my favorite Jackbox game of all time. I've laughed my ass off at many shirts. It's a little slow, and it could use a better prompt system, but when it gets going it is absolutely worth it.
Champ'd Up from Party Pack 7 is a spiritual successor, but the concept and voting system are a little different. I prefer Tee K.O. personally, but Champ'd Up is good too. Plus it has a much better drawing system, with separate lineart and colors. Look into whether you'd like to play that instead.
---
FAKIN' IT
Everyone gets a question. One person gets a fake question, and has to blend in. Everyone else has to find the faker.
I've never played this myself, because it requires playing IRL. There are prompts that ask you to make faces, or point at other people in the room, or hold up numbers on your hands. Videos I've seen on it have been hilarious, but be aware that if you're streaming over Discord or something, you probably won't be playing this unless you develop some sort of workaround.

If you only want to get one Jackbox pack, 3 is probably your best bet.
+Tee K.O. (5) - Drawing, writing, and matching game where players are competing to make the best (funniest) t-shirt designs using a pool of everyone's drawing and writing submissions. Only downside is it takes much longer to play than other Jackbox games.
+Quiplash 2 (4) - Sequel to the writing prompt competition game.
+Fakin' It (4) - Great social deduction party game, but only playable if all players are physically present.
+Guesspionage (3) - Number-based trivia guessing game based on results of rather unscientific polls.
+Trivia Murder Party (2*) - Horror-themed trivia game. I know a lot of people like this one, but I felt it was really unbalanced and oddly paced. These issues seem to be rectified in its sequel.

Individual reviews (one star for an enjoyable game, half for semi)
Quiplash 2: It's quiplash, what else is there to say.
Guesspionage: Honestly is a little underrated. Guess percentages is honestly fun and seeing how wrong you are just shocks and gets everyone going "how"
Trivia Murderer Party: TriviaXMini-Games is genius
Fakin It: Need to be in-person to play it, but still really fun just to see it all go down, as long as everyone is able to understand the rules
TKO: The best drawing game in Jackbox, you draw and you write and you get others dumpster fires to make a bigger dumpster fire and fight against others dumpster fires...it's honestly really fun
